The 27-year-old Irishwoman emerged as a star when helping Team Europe win the Solheim Cup in 2021.
Maguire added to the Solheim Cup victory as she won the Drive On Championship by three shots from Lexi Thompson at Crown Colony Golf & Country Club in Fort Myers, Florida.
Maguire held the overnight lead with Marina Alex, and her five-under par final round was enough for a comfortable victory in what was her 50th LPGA start.
It was a third professional win for Maguire, who previously spent a year as the number one ranked amateur in the game.
Maguire won twice on the Symetra Tour in 2019 with victories in the Windsor Golf Classic and the Symetra Classic.
